Myh Deficiency Enhances Intestinal Tumorigenesis in Multiple Intestinal Neoplasia (Apc ) Mice
نویسندگان
چکیده
Monoallelic APC and biallelic MYH (homolog of Escherichia coli mutY) germ-line mutations are independently associated with a strong predisposition to colorectal adenomas and carcinoma in humans. Whereas mice heterozygous for mutant Apc develop intestinal tumors, mice homozygous for mutant Myh do not show increased tumor susceptibility. We analyzed the phenotype of Apc /Myh / mice and found that they developed significantly more adenomas in the small intestine than did Apc / Myh / or Apc /Myh / mice (median 231 versus 151 versus 152). In the large bowel, Apc /Myh / mice showed significant increases in the number of aberrant crypt foci. In addition, Apc /Myh / mice developed an increased number of mammary tumors. Molecular analyses suggested that at least 19% of intestinal tumors from Apc /Myh / mice had acquired intragenic Apc mutations rather than allelic loss. Consistent with a defect in base excision repair, three intragenic Apc mutations in polyps without allelic loss from Apc /Myh / mice were shown to be G:C to T:A transversions which resulted in termination codons; no such mutations were found in polyps from Apc /Myh / or Apc /Myh / mice. Tumors from Apc /Myh / mice harbored neither somatic mutations nor allelic loss at Myh. Thus, homozygous, but not heterozygous, Myh deficiency enhanced intestinal tumorigenesis in Apc mice. The excess small-bowel adenomas in Apc /Myh / mice, therefore, appear to be a model of MYH-associated polyposis in humans.

متن کاملGenetic and Diet-Induced Obesity Increased Intestinal Tumorigenesis in the Double Mutant Mouse Model Multiple Intestinal Neoplasia X Obese via Disturbed Glucose Regulation and Inflammation
We have studied how spontaneous or carcinogen-induced intestinal tumorigenesis was affected by genetic or diet-induced obesity in C57BL/6J-Apc (Min/+) X C57BL/6J-Lep (ob/+) mice. Obesity was induced by the obese (ob) mutation in the lep gene coding for the hormone leptin, or by a 45% fat diet. The effects of obesity were examined on spontaneous intestinal tumors caused by the multiple intestina...
متن کاملThe Distinct Spectra of Tumor-associated Apc Mutations in Mismatch Repair-deficient Apc Mice Define the Roles of MSH3 and MSH6 in DNA Repair and Intestinal Tumorigenesis
In mammalian cells, mismatch recognition has been attributed to two partially redundant heterodimeric protein complexes of MutS homologues, MSH2-MSH3 and MSH2-MSH6. We have conducted a comparative analysis of Msh3 and Msh6 deficiency in mouse intestinal tumorigenesis by generating Apc mice deficient in Msh3, Msh6 or both. We have found that Apc mice defective in Msh6 show reduced survival and a...
متن کاملDisruption of estrogen receptor signaling enhances intestinal neoplasia in Apc(Min/+) mice.
Estrogen receptors (ERs) [ERalpha (Esr1) and ERbeta (Esr2)] are expressed in the human colon, but during the multistep process of colorectal carcinogenesis, expression of both ERalpha and ERbeta is lost, suggesting that loss of ER function might promote colorectal carcinogenesis. Through crosses between an ERalpha knockout and Apc(Min) mouse strains, we demonstrate that ERalpha deficiency is as...
متن کاملErythrocyte polyamine levels are greater than triglyceride levels as markers of intestinal polyp formation in Min mice
Multiple intestinal neoplasia (Min) mice were originally identified by Moser et al. Min mice have a heterozygous mutation in the tumor suppressor gene, adenomatous polyposis coli (Apc). The APC gene is mutated in familial adenomatous polyposis (FAP). Min mice spontaneously develop intestinal polyps, similar to humans, but mainly in the small intestine. Polyps form as early as approximately 4 we...
